Unitary sodium channels in isolated cardiac myocytes of rabbit.

چکیده
Transmembrane ionic movements are thought to occur through special membrane areas or channels. Until recently, it has not been possible to study the properties of individual ionic channels, directly. We have used the extracellular patch clamp technique to resolve unitary sodium channels in enzyme-dissociated rabbit ventricular myocytes. Depolarizing voltage clamp steps elicited rectangular inward current fluctuations representing open-closed transition of the channels. Probability of channel opening increased with depolarization. Single channel current amplitude ranged from 0.6 to 1.9 pA. Channel open time increased with depolarization. Sodium channels could be distinguished from calcium channels by their insensitivity to cadmium and inactivation voltage range. First-event latency histograms suggest multiple-closed states. The patch clamp technique can be readily applied to the study of sodium channels in adult cardiac myocytes.

Effect of Sodium Valproate on Ouabain-Induced Arrhythmia in Isolated Guinea-Pig Atria
Sodium valproate (SV), an antiepileptic drug has several mechanism of action. It inhibits voltage sensitive Na+ channels and reduces intracellular Na accumulation. These actions are similar to that of both phenytoin and carbamazepine. We have investigated the direct cardiac action of SV and its effects on ouabain-induced arrhythmia in isolated guinea-pig atria. The guinea-pig atrium was dissect...

To investigate possible ionic current mechanisms underlying ischemic arrhythmias, we studied single Na+ channel currents in rat and rabbit cardiac myocytes treated with the ischemic metabolite lysophosphatidylcholine (LPC) using the cell-attached and excised inside-out patch-clamp technique at 22 degrees C. LPC has been reported previously to reduce open probability and to induce sustained open...
متن کاملIntracellular calcium modulation of voltage-gated sodium channels in ventricular myocytes.
AIMS Cardiac voltage-gated sodium channels control action potential (AP) upstroke and cell excitability. Intracellular calcium (Ca(i)(2+)) regulates AP properties by modulating various ion channels. Whether Ca(i)(2+) modulates sodium channels in ventricular myocytes is unresolved. We studied whether Ca(i)(2+) modulates sodium channels in ventricular myocytes at Ca(i)(2+) concentrations ([Ca(i)(...
متن کاملIntegrative Physiology Blocking Scn10a Channels in Heart Reduces Late Sodium Current and Is Antiarrhythmic
Rationale: Although the sodium channel locus SCN10A has been implicated by genome-wide association studies as a modulator of cardiac electrophysiology, the role of its gene product Nav1.8 as a modulator of cardiac ion currents is unknown. Objective: We determined the electrophysiological and pharmacological properties of Nav1.8 in heterologous cell systems and assessed the antiarrhythmic effect...
